چکیده
While a significant number of studies have focused on elucidating the functioning mechanisms Hepatocellular carcinoma (HCC) microenvironment, intercellular crosstalk between multiple cells in tumor microenvironment remains unclear. Here we co-cultured spheroids HCC cells, hepatic stellate (HSCs), and hepatocytes biomimetic composite hydrogel to construct 3D model vitro. The reproduced major cellular components early realizing visualization interplay microenvironment. Using this model, showed that HSCs were activated when with deposed collagen remodel which turn triggered higher EMT levels cells. also responded existence activation co-culture, showing downregulated expression level ALB, AFP, HNF4A. This recapitulated enabled multicellular 3D, providing platform investigate related fibrosis.

Intercellular Adhesive Structures Between Stellate Cells – An Analysis in Cultured Human Hepatic Stellate Cells
To investigate whether or not hepatic stellate cells can form intercellular junctions with each other, we cultured human stellate cells (LI90) on different kinds of substrata. Intercellular junctions were detected between these cultured stellate cells by transmission electron microscopy (TEM). The molecular components of the intercellular adhesive structures were identified by immunofluorescenc...

Herein, we review the current findings of how a variety of accessory cells could participate in shaping the tumor microenvironment and supporting the mechanisms by which cancer cells undertake the epithelial-mesenchymal transition (EMT). EMT, a complex of phenotypic changes, promotes cancer cell invasion and creates resistance to chemotherapies. Among the accessory cells present in the EMT, imm...

متن کاملTIMP-1 mediates TGF-β-dependent crosstalk between hepatic stellate and cancer cells via FAK signaling
Transforming growth factor-β (TGF-β) signaling plays a key role in progression and metastasis of HCC. This study was undertaken to gain the proof of concept of a small-molecule inhibitor of TGF-β type I receptor kinase, EW-7197 as a potent anti-cancer therapy for HCC. We identified tissue inhibitors of metalloproteinases-1 (TIMP-1) as one of the secreted proteins of hepatic stellate cells (HSCs...
